Readily available and inexpensive technologies could double or triple fuel efficiency

It has been half a century since the truck industry started experimenting with more efficient designs, and steady improvements to engines and modest aerodynamic advances have followed in the years since. Unfortunately, the gains have been undone by other factors that have increased fuel consumption, such as higher speeds, traffic congestion and excessive product packaging, which has cut the effective capacity of each truck. Meanwhile the cargo trailer - the box-on-wheels that is pulled by the tractor - has been virtually ignored. "It's a dumb asset, and it has to get smarter than that," says Bill Van Amburg of Calstart, a low-energy-transportation consultancy in Pasadena, California.

Yet there are ways of streamlining trailers. Skirts that cover the wheels and the open space under the trailer, and fairings covering the gap between the tractor and trailer, can improve fuel efficiency by 6 per cent. Rounded flaps or fins on the back of the trailer add a further 6 per cent. And simply blowing air through vents at the rear of the trailer cuts drag by up to 5 per cent (see "Go with the flow"). Wind-tunnel tests confirmed many of these improvements more than half a century ago. So why are they not commonplace?

Part of the problem is that with about four trailers to each tractor in the US, the trailers sit idle most of the time, reducing the payback from any investment aimed at enhancing fuel efficiency. To make matters worse, it's the company operating the tractor unit that pays the fuel bill, while the trailer may well be owned by someone else. "It's really a battle between logistics, practicality and cost," says Ken Howden, director of the 21st Century Truck Partnership, a project to improve truck performance coordinated by the US Department of Energy.The US is the world's largest producer and exporter of crops, accounting for around 40 per cent of global maize and soybean production.

"If US yields go down a lot, it could drive up prices of staple food commodities all around the world", say the researchers. "Almost surely the poor would suffer far more that the US would."

David Pimentel at Cornell University in Ithaca, New York, argues that curbing biofuel production would tackle starvation and high food prices far quicker than curbing greenhouse gas emissions. "Some 66 per cent of the world population malnourished, yet the US turns 33 per cent of its corn crops into biofuels," he says.The idea of using CO2 as a carbon source is appealing," says Philip Jessop, a chemist at Queen's University in Kingston, Ontario, Canada.

But to make a jet fuel that is properly "green", the energy-intensive electrolysis that produces the hydrogen will need to use a carbon-neutral energy source; and the complex multi-step process will always consume significantly more energy than the fuel it produces could yield. It uses a variant of a chemical reaction called the Fischer-Tropsch process, which is used commercially to produce a gasoline-like hydrocarbon fuel from syngas, a mixture of carbon monoxide and hydrogen often derived from coal.

Robert Dorner, a Naval Research Laboratory chemist in Washington DC and first author of a new paper on the technique, says that CO2 is rarely used in the Fischer-Tropsch process because of its chemical stability.

But CO2's abundance, combined with concerns about global warming, make it an attractive potential feedstock, Dorner says. Although the gas forms only a small proportion of air – around 0.04 per cent – ocean water contains about 140 times that concentration, he says.
Iron catalyst

The navy team have been experimenting to find out how to steer the CO2-producing process away from producing unwanted methane to produce more of the hydrocarbons wanted.

In the conventional Fischer-Tropsch process, carbon monoxide and hydrogen are heated in the presence of a catalyst to initiate a complex chain of reactions that produce a mixture of methane, waxes and liquid fuel compounds.

Dorner and colleagues found that using the usual cobalt-based catalyst on seawater-derived CO2 produced almost entirely methane gas. Switching to an iron catalyst resulted in only 30 per cent methane being produced, with the remainder short-chain hydrocarbons that could be refined into jet fuel.While the average fuel efficiency of the US car fleet has almost doubled in the last 40 years, today's heavy trucks guzzle the same amount of fuel - roughly 30 litres per 100 kilometres - as they did in 1969 (see graph). In 1990, America's truckers burned the equivalent of 1.6 million barrels (254 million litres) of oil per day, about 10 per cent of the nation's total consumption. By 2007, this had risen to 2.5 million barrels.

This cannot go on, and truck-makers, freight companies and transport researchers are finally getting the message. Across the world, and especially in the US, they are taking a new look at ways to cut freight fleets' fuel consumption.

Even small improvements can bring big benefits. A concept vehicle called the Innovation Truck unveiled in April by Daimler Trucks North America is designed to show what is possible by tweaking a vehicle's aerodynamics. In wind tunnel tests it used 3 per cent less fuel than an unmodified truck - which rolled out across the entire US truck fleet would save about 4.3 billion litres of fuel annually. This translates into an annual cut in CO2 emissions of over 11 million tonnes.

More ambitious plans are afoot. Engineers and industry leaders at an RMI conference on freight-vehicle efficiency in April concluded that readily available and inexpensive technologies could boost efficiency by up to 23 per cent, and even doubling or tripling fuel efficiency might be within reach.

That's likely to rise as the world warms. Even if we reduce emissions by 50 per cent by 2050 relative to 1991 levels – a target that governments are struggling to agree on – yields could still fall by between 30 and 46 per cent.

"These estimates of yield loss are amongst the best we have," says David Bohan, head of the Ecosystem Dynamics and Biodiversity group at Rothamsted Research, UK.Even densely populated regions like southeast Asia typically have more than a third of the farmland covered in trees, says author Robert Zomer.

"Pioneer farmers remove trees from the landscape, but as intensive systems develop, there is an increase in planting useful trees," says Zomer.

Navy chemists have processed seawater into unsaturated short-chain hydrocarbons that with further refining could be made into kerosene-based jet fuel. But they will have to find a clean energy source to power the reactions if the end product is to be carbon neutral.

The process involves extracting carbon dioxide dissolved in the water and combining it with hydrogen – obtained by splitting water molecules using electricity – to make a hydrocarbon fuel.

Screening for the genetic fragments could act as an early warning system for beekeepers, says Berenbaum. "This could open a new avenue for colony monitoring," says Peter Neumann at the Swiss Bee Research Centre in Switzerland.Farmers wreck forests, right? You would be hard-pressed to find anyone who disagreed, but it turns out past studies have seriously under-estimated the number of trees on farms around the world.

In fact, a new analysis by the World Agroforestry Centre in Nairobi (pdf) shows that often, the more intensive the farming the more trees that farmers plant – for fruit, medicines, fodder crops, windbreaks, and fuel.

The study, presented at the World Congress of Agroforestry in Nairobi this week, uses satellite images to show that almost half of farmed land worldwide has significant tree cover, most of it previously unmapped.

Of the 22 million square kilometres of farmland around the world about half had at least 10 per cent tree cover. About 7 per cent of land classified as agricultural had more than 50 per cent tree covWhile news reports and disaster movies remind us about tipping points for Arctic melt and sea level rise, some things closer to home get less attention. Take food supply: new modelling studies show that there are climate tipping points here too, beyond which crop yields will collapse.

Wolfram Schlenker at Columbia University, New York, and Michael Roberts at North Carolina State University in Raleigh used a high-resolution dataset of weather patterns from 1950 to 2005 to discover how yields of three key US crops would respond to increasing temperatures.

"The single best predictor of a year's yield is the amount of time temperatures exceed about 29 °C and the extent to which they do so," they say.

"Below this, warmer temperatures are beneficial for yields, but the damaging effects above 29 °C are staggeringly large."The results were striking. Red-headed birds won the contests every time, regardless of their inherited colour or the upbringing of both combatants. "Painting the birds did not make them act more aggressively, but they won the food contests because the other birds wouldn't challenge them at the feeder," Pryke reports. "All the birds reacted strongly to red-painted birds - even ones that had never seen a red-headed bird before."

After each interaction, Pryke measured levels of the hormone corticosterone, a reliable measure of stress. She found that birds facing a red opponent had 58 per cent higher corticosterone levels than birds that faced black or blue. "With no prior experience of the colour, these birds had an innate fear of red," Pryke concludes.

Exposure to red doesn't just have an effect on fights. In a remarkable series of studies, Andrew Elliot of the University of Rochester in New York has demonstrated that even a brief glimpse of red can change human abilities and behaviour in all sorts of ways.

In one experiment, volunteers were asked to carry out a 5-minute IQ test. They were assigned a bogus "participant number", written in either red or black, on the corner of the test paper. Volunteers whose numbers were written in red scored consistently lower on the tests. Recent evidence supports the idea that red exerts its effect on humans via perceptions of dominance. In an experiment, Hill and colleague Tony Little showed 105 volunteers different coloured circles and asked them to indicate which would be "most likely to win a physical competition" and which circle looked "most dominant". Red won hands down (Journal of Evolutionary Psychology, vol 5, p 161).

Is this response to red innate or learned? Again, animal experiments give some clues. In July, Sarah Pryke at Macquarie University in Sydney, Australia, published the results of a study into Gouldian finches, which, depending on their genes, have either red or black head feathers. "Red-headed birds dominate black-headed ones," Pryke says. "I wanted to find out whether this results from nature or nurture."

Pryke raised newly hatched chicks with red or black heads in different family groups, so that they grew up in isolation or with adults of both head colours. She now had birds with black or red heads and various experience of colour. Next, to test what difference their experience of colour made, Pryke painted the juveniles' heads either red, black or blue (as a control) and put them into 20-minute contests, where two birds would compete for space at a bird feeder (Animal Behaviour, vol 78, p 393).Storms on the move

Existing forecasting computer systems use broad-brush methods: for instance, trying to predict overall stormy conditions over a region. They show a "threat area" that thunderstorms are likely to pass through.

Like all weather forecasts they are not completely reliable, but crucially they do not tell forecasters how far wrong the prediction is likely to be. So the forecasters must take the predicted threat area and draw in, by hand, a slightly wider region. They then issue a thunderstorm warning for this larger area.

THESPA's use of strike probabilities removes this element of guesswork: it detects individual storms and predicts their paths. As a result, Dance says, "Our system is more geographically specific."
Thundering good

The system was "trained" using records of thunderstorms near Sydney, Australia. It was then tested on a database of 2394 storm tracks, which were recorded in the Beijing area of China around the time of the 2008 Olympics.

The system performed well, achieving a Brier Skill Score of 0.39. On this scale, 1 would be a perfect score, and 0 would be as good as the present manual forecasting method.Beekeepers have seen hive after hive fall prey to colony collapse disorder (CCD). Now insights from the honeybee genome could overthrow guesswork in the effort to diagnose the cause of the die-offs.

May Berenbaum at the University of Illinois, Urbana-Champaign, and her colleagues looked for genetic differences between bees from US colonies that have suffered CCD and bees that were sampled before colony collapses shot up in 2006. CCD killed off about a third of US honeybees in 2007 and 2008.

The team found 65 genes that were distinctly different in CCD bees. They also discovered unusual snippets of genetic material that are typical of infection with the RNA viruses known as picorna-like viruses. They found no evidence to suggest that pesticides or bacterial infection are the primary cause of CCD. Berenbaum thinks picorna-like viruses may be the root cause, making the bees highly vulnerable to other viruses, pesticides and bacteria.

Why Obama's Dog Has Curly Hair

ScienceDaily (Aug. 28, 2009) — University of Utah researchers used data from Portuguese water dogs – the breed of President Barack Obama's dog Bo – to help find a gene that gives some dogs curly hair and others long, wavy hair.

It was part of a National Institutes of Health (NIH) study – published online Thursday, Aug. 27 by the journal Science – showing that variations in only three genes account for the seven major types of coat seen in purebred dogs. The findings also point the way toward understanding complex human diseases caused by multiple genes.

"We were part of a team that found three genes that control 90 percent of the seven coat types that characterize different breeds of purebred dogs," says K. Gordon Lark, one of 20 co-authors of the study and a distinguished professor emeritus of biology at the University of Utah.

"We helped identify the gene that controls curly or wavy coats," adds study co-author Kevin Chase, a University of Utah research specialist in biology.

The study's first author is Edouard Cadieu, and its senior author is Elaine Ostrander, both of the National Human Genome Research Institute in Bethesda, Md. Other authors came from that institute, which is part of NIH, and from the University of California's Davis and Los Angeles campuses, Cornell University in Ithaca, N.Y., and Affymetrix Corp., in Santa Clara, Calif.

The study showed that combinations of various forms of only three genes – named RSPO2, FGF5 and KRT71 – account for seven major coat types in purebred dogs.

By analyzing more than 1,000 dogs from 80 domestic breeds, the researchers found that RSPO2 is the gene associated with whether or not a dog has a moustache and large eyebrows (known together as "furnishings"); FGF5 is linked to whether a dog's fur is long or short; and KRT71 determines if the hair is curly or wavy.

Mutant Genes and the Well-Dressed Dog

Artificial selection, at the heart of breeding for desirable traits in domesticated animals, has yielded rapid change in a short span of canine history. While researchers estimate that modern dog breeds diverged from wolves some 15,000 years ago, the genetic changes in the dog genome that create multiple coat types are more likely to have been pursued by breeders in just the past 200 years. In fact, breeds with short, straight hair, such as the beagle, display the original, more wolf-like versions of the three genes identified in the study.

Modern dog breeds are part of a unique population structure, having been selectively bred for many years. Based on this structure, the researchers were able to break down a complex phenotype — coat — into possible genetic variations. "When we put these genetic variants back together in different combinations, we found that we could create most of the coat varieties seen in what is among the most diverse species in the world — the dog," Dr. Ostrander said. "If we can decipher the genetic basis for a complex trait such as the dog's coat, we believe that we can do it as well with complex diseases."

All purebred dogs have the three genes, but the presence or absence of mutant, variant forms of those genes – rather than the ancestral forms inherited from wolves – determines coat types:

* Short-haired dogs like basset hounds have none of the variant genes, just the ancestral form of each gene.
* Wire-haired dogs such as Australian terriers have the variant form of only the RSPO2 gene.
* Dogs with wiry and curly hair – airedale terriers, for example – have variants of both RSPO2 and KRT71 genes.
* Long-haired dogs like golden retrievers have a variant form of the FGF5 gene.
* Long-haired dogs with furnishings, such as the bearded collie, have variant forms of FGF5 and RSPO2.
* Curly haired dogs such as Irish water spaniels have the variant forms of the FGF5 and KRT71 genes.
* Curly haired dogs with furnishings – such as the bichon frise breed and some Portuguese water dogs, including President Obama's dog Bo – have the variant form of all three genes.

Portuguese water dogs come in both curly haired and wavy haired varieties, depending on what form of the KRT71 gene they have. Chase says wavy haired Portuguese water dogs fit in the new study's long hair with furnishings category.

Chase says breeders characterized First Dog Bo as curly with furnishings, while Lark's Portuguese water dog, Mopsa, has wavy hair, even though she appears pretty curly. Chase notes that the KRT71 gene really controls the extent of curl, not curl versus no curl

What about the fur characteristics of mutts? Because mutts are not part of a closed breeding population like each breed of purebred dog, it's difficult to separate the genetic contributors to their traits, Chase says.

"We don't know enough about the genetics of mutts," Lark adds.

From Portuguese Water Dogs to Human Disease

Chase and Lark assisted the NIH research by sharing data on Portuguese water dogs, which they have studied for years.

"Our contribution to this paper is data collected on the Portuguese water dog that indicated that keratin genes controlled the amount of curliness," Lark says. "Our data plus other data from the Ostrander group narrowed this down to KRT71."

KRT71 carries the code that produces keratin 71, a structural protein in hair.

Even though he and Chase studied the genetics of curliness stemming from KRT71, Lark says the other two genes involved in determining dog coat types are more interesting in the sense that they produce proteins that regulate a variety of processes in living organisms, not just the kind of coat a dog has. That makes them relevant to diseases of dogs and humans.

"Dogs share many diseases and other traits with humans," Lark says. "That's why they were used for many decades so extensively for pharmaceutical and medical-physiological-biochemical testing. Not surprisingly they share much of their genome [genetic blueprint] with humans also."

Lark says a dog bred for a desired trait may end up, years later, more vulnerable to cancer, immune disorders or other diseases of aging, and thus have a shorter life span.

"Ultimately the most important part of this paper – mostly the work of Ostrander's group – is that we will be able to get glimpses of how major regulatory genes interact with other genes to change the functioning of an animal in a way that does not kill it, but that may eventually compromise its longevity or its functioning as it ages," Lark says.

Earlier studies involving Ostrander and Lark revealed genetic variations that make small dogs small and account for longevity in dogs.

A Pup for a President and a Geneticist

Lark, once a soybean geneticist, has run a long-term dog genetics research effort known as the Georgie Project, after Georgie, a stray Portuguese water dog he adopted in 1986. Georgie died in 1996, and when Lark sought a replacement, a breeder sent him Mopsa – now 13 years old – and nagged him to study the breed's genetics.

Lark and Chase learned that the breed was ideal for genetics research because all Portuguese water dogs descended from a small group of "founders" and could help the search for genes responsible for complex traits – including human diseases caused by multiple interacting genes.

Portuguese water dogs gained publicity earlier this year when President Obama fulfilled his promise to get a dog for his daughters by obtaining Bo, a Portuguese water dog, reportedly as a gift from U.S. Sen. Edward Kennedy, D-Mass.

"As a supporter of Obama, I feel glad that he has a good dog," Lark says. "He has good taste and a good dog."

Finnish Scientists Discover Nerve Growth Factor With Therapeutic Potential In Parkinson's Disease

ScienceDaily (Aug. 30, 2009) — Scientists in the Academy of Finland's Neuroscience Research Programme have reported promising new results with potential implications for the treatment of Parkinson's disease. They have been studying the impacts of nerve growth factors in the treatment of PD, and their latest results show that a certain growth factor can be used to halt the progress of damage brought on by a nerve poison and possibly even restore the function of damaged cells.

The studies on nerve growth factors used an experimental PD model in rats. Administration of the growth factor reduced motor disturbances in rats.

The severe motor disturbances that are seen in PD are caused by the slow degeneration of dopamine nerves in the brain. There are treatments that alleviate the symptoms of the disease, such as hand tremor, but they do not prevent or halt the degeneration of nerve cells. The nerve growth factors studied to date have slowed nerve cell degeneration to some extent, but they have had only limited therapeutic effect. Several known nerve growth factors, such as GDNF, also attach to extracellular tissue, possibly deterring their movement to nerve cells that require treatment.

Working under the supervision of Academy Professor Mart Saarma, scientists at the University of Helsinki Institute of Biotechnology have now been investigating two new nerve growth factors. MANF (mesencephalic astrocyte-derived neurotrophic factor) is released from glial cells in the midbrain and is a member of the same growth factor family as CDNF, another growth factor that Saarma's team have investigated. A University of Helsinki team led by Professor Raimo K. Tuominen discovered that in the experimental PD model, MANF and CDNF injections into the brain prevented dopamine nerve destruction caused by nerve poison and to some extent even restored the function of damaged cells in rats.

The latest results suggest that MANF spreads more readily in brain tissue than other known growth factors. This may be a highly significant finding in respect to the development of growth factor therapy for PD.

The results are published in the 29 July issue of the Journal of Neuroscience.

'Curtain Twitching' Skylarks Keep Track Of Strangers Through Their Songs

ScienceDaily (Aug. 30, 2009) — Skylarks can hear the difference between friendly neighbours and dangerous strangers, and deal with any threatening intruders, says new research by scientists at Queen Mary, University of London.

Male skylarks learn to recognise local dialects in their neighbours' individual songs, remember where each neighbour is supposed to be and reprimand intruders who don't belong in the neighbourhood, according to a study carried out by Dr. Elodie Briefer, a postdoctoral researcher at Queen Mary's School of Biological and Chemical Sciences.

Published in the Springer journal Naturwissenschaften ¹ this week, Dr. Briefer and her colleagues at the University of Paris South found that skylark neighbours are tolerated if they stay in their own territory, whereas strangers - skylarks who belong to another neighbourhood - are attacked if they intrude too close to the nest.

Researchers also observed the birds' reactions when they heard the recorded song of another skylark from different directions. The results of the study showed how neighbouring birds who travel too far from their regular territory - a move which is seen as threatening - also run the risk of being attacked.

Males skylarks fiercely guard their chosen home territory, the area of land where they make their nest and hunt for food. The size and position of the male's territory is also important as female birds check it out before deciding who is going to make the best father to her chicks. Each skylark will usually have several neighbours, living in territories that border his own.

Bird songs are among the most complex sounds produced by animals and the skylark (Alauda arvensis) is one of the most complex of all. The songs are composed of 'syllables', consecutive sounds produced in a complex way, with almost no repetition. The male skylark can sing more than 300 different syllables, and each individual bird's song is slightly different.

Dr. Briefer's research found that the songs of neighbouring skylarks share more syllables with each other than they do with strangers, like a dialect. She says: "This may have evolved because it is safer for the birds to live close together, but they need a way to keep intruders out. By sharing a local dialect in their song, they can keep an ear out for other birds that live nearby and kick any strangers out of the neighbourhood."

Novel Genetic Region Identified For Childhood Asthma In Mexicans

ScienceDaily (Aug. 30, 2009) — Genetic variants in a region on chromosome 9q may influence asthma development in Mexican children, according to research published in the August 28 issue of the open-access journal PLoS Genetics.

Researchers from the National Institute of Environmental Health Sciences and the National Institute of Public Health (Mexico) and their collaborators at universities in the US and the UK conducted a genome-wide association (GWA) study, in which they looked at over 500,000 variants across the genome in 492 Mexican children with asthma and their parents to identify novel genes that may influence asthma development. The work points to the chromosome 9q21.31 region as a novel candidate region for childhood asthma.

Asthma is a leading chronic childhood disease that is influenced by both genetic and environmental factors. However, few genes have been consistently associated with the disease. GWA studies have successfully identified novel genes for many common diseases, but to date there have been only a handful of GWA studies focused on asthma and even fewer focused on asthma in Hispanic populations.

The researchers also examined ancestry in this Mexican population and found that the chromosome 9q21.31 region may underlie some of the differences in childhood asthma prevalence that have been observed across ethnic groups. It remains unclear why Mexicans have lower rates of asthma than some other groups.

The chromosome 9q21.31 variants associated with childhood asthma in this study are located near the TLE4 gene, but the researchers state that "further work is needed to decipher whether TLE4 or a nearby gene explains the signals from the chromosome 9q21.31 region." In addition, it is likely that multiple genetic and environmental risk factors underlie the development of childhood asthma, and the researchers conclude that studies with very large sample sizes will be needed to identify the important interacting risk factors.

Scientists Hone Technique To Safeguard Water Supplies

ScienceDaily (Aug. 30, 2009) — A method to detect contaminants in municipal water supplies has undergone further refinements by two Oak Ridge National Laboratory researchers whose findings are published on line in Water Environment Research.

The new work demonstrates that the technology that uses algae as sentinels has broader applications than previously reported, according to authors Miguel Rodriguez Jr. and Elias Greenbaum of the Department of Energy's ORNL. For example, under real-world operating conditions, the sensitivity of the algae to toxins has a natural daily cycle that tracks the sun.

"When the sun is overhead and shining brightly, the algae are less sensitive to the toxins," Greenbaum said. "The new work shows that keeping the water sample in darkness for about 30 minutes prior to testing for toxins restores full sensitivity to the test."

The new results also show that the technology can be applied to many different water quality environments such as when the algae are starved for nutrients.

"Our key result is that despite real-world conditions that create challenges, free-living microalgae combined with 'work-around' strategies can be used as broad-spectrum automated biosensor systems for continuous monitoring of source drinking water," Greenbaum said.

The process uses a fluorometer to measure the fluorescence signal of algae that grow naturally in source water such as Tennessee's Clinch River, which was used in this study. Researchers exploit the known characteristics of Photosystems I and II, which convert light energy into chemical energy, to detect any changes in the process of photosynthesis.

"Recent advances in optoelectronics and portability make this a powerful technology for monitoring the in situ physiology of aquatic photosynthetic organisms such as green algae and cyanobacteria," the authors wrote. Even low levels of toxins alter fluorescence patterns within minutes.

Another significant aspect of this work is the reporting of statistically reliable data on the threshold detection levels for broad classes of toxins such as blood and nerve agents and agrochemicals. These levels are at or near Environmental Protection Agency regulatory guidelines, Greenbaum said.

For this study, the researchers looked at five classes of chemical agents in water: Diuron, atrazine, paraquat, methyl parathion and potassium cyanide. All are known to be harmful to human health. In the case of Diuron, used in agriculture for 50 years, Greenbaum and Rodriguez were able to detect 1 part per million. This was indicated by a 17 percent decline in the algae's Photosystem II efficiency.

"We have shown that microalgae in source drinking water can be used as broad-spectrum, robust sentinel sensors to detect relatively low concentrations of toxins," Greenbaum said. "We have also shown that the microalgae do not need to be in an optimized state for this technology to be effective."

This research was funded by the Department of Energy's Office of Biological and Environmental Research, the Defense Advanced Research Projects Agency and BAE Systems. Discussions for commercialization of this technology, to be marketed under the name AquaSentinel, are under way.

Familiar And Newly Learned Words Are Processed By The Same Neural Networks In The Brain

ScienceDaily (Aug. 30, 2009) — Our vocabulary continues to grow and expand even in adulthood. Just ten years ago, the word 'blog' did not yet exist – and now we no longer remember when we heard this word for the first time or when we learned its meaning. At some stage new words become just as familiar to us as words we have learned earlier. One of the areas of interest in the Academy of Finland's Neuroscience Research Programme (NEURO) is how the process of learning new words is reflected in the function of the brain.

The new research evidence emerging about how the brain processes language and its different levels has important application among other things in the development of language teaching.

In one of the experiments conducted in the NEURO programme, participants learned the name and/or purpose of 150 ancient tools. They had never heard these words before. The subjects' brain function was measured by means of magnetoencelography during the naming of the tools, both before and after the learning period.

The results show that the brain uses the same neural networks to process both familiar and newly learned words. The names of objects were processed in the left temporal and frontal lobe within half a second of showing the image of the tool to the subject. "If the subject had only recently learned the name of the tool, the the naming process induced an activation that was just as strong or stronger than the activation induced by the image of a familiar object," says Academy Professor Riitta Salmelin, HUT Low Temperature Laboratory, who is in charge of the research.

According to Salmelin the learning of the meaning of ancient tools did not cause corresponding clear differences in the function of the brain. In other words, it seems that the processing of meanings in the brain differs essentially from the processing of names. On the other hand, the performance results indicated that new definitions were learned even faster than new names.

How are learned words retained?

The research team are now working on a follow-up study to explore the retention of learned words. Is it possible to detect in brain activation some specific phenomenon that predicts good retention of learned knowledge up to several months after learning?

"We are also conducting a separate series of experiments to find out how our brain learns phonetic structures and, on the other hand, how the brain learns to identify letter combinations that are typical of a certain language," Riitta Salmelin explains.

Another area of interest in the ongoing study is the role of grammar in language learning. The focus here is to explore how the brain learns to use the vocabulary and grammatical structure of an experimental miniature language.

The research project is a joint undertaking of the HUT Low Temperature Laboratory, the Department of Psychology at Åbo Akademi University and the University of Helsinki Department of Psychology.

Scientists Construct 'Off Switch' For Parkinson Therapy

ScienceDaily (Aug. 29, 2009) — A common antibiotic can function as an "off switch" for a gene therapy being developed for Parkinson's disease, according to University of Florida researchers writing online in advance of September's Molecular Therapy.

The discovery in rats answers an important question — how can new, therapeutic genes that have been irrevocably delivered to the human brain to treat Parkinson's be controlled if the genes unexpectedly start causing problems?

Meanwhile, in a review of Parkinson treatments, the researchers say that prior experimental attempts using growth factors — naturally occurring substances that cause cells to grow and divide — to rescue dying brain cells may have failed because they occurred too late in the course of the disease.

Together, the findings suggest that gene therapy to enable the brain to retain its ability to produce dopamine, a neurotransmitter that falls in critically short supply in Parkinson's patients, could be safely attempted during earlier stages of the disease with an added likelihood of success.

Parkinson's disease affects more than 1 million Americans, causing patients to gradually develop movement problems, including tremors, stiffness and slowness. It is caused by degeneration and death of nerve connections that produce dopamine, a substance necessary for communication between cells that coordinate movement.

"We have worked every day for 10 years to design a construct to the gene delivery vector that enhances the safety profile of gene transfer for Parkinson's disease," said Ronald Mandel, a professor of neuroscience at UF's McKnight Brain Institute and the Powell Gene Therapy Center. "With that added measure of safety, we believe we can intervene with gene transfer in patients at earlier stages of the disease. We strongly believe that trials to save dopamine-producing connections in patients with Parkinson's disease have failed because the therapy went into patients who were in the late stages of the disease and who had too few remaining dopamine-producing connections."

Often patients are given prescriptions for levodopa, or L-dopa, which is converted into dopamine by enzymes in the brain. But the treatment loses its effectiveness over time and does nothing to slow the disease's progression.

Meanwhile, trials in the United States to treat Parkinson's involving direct infusion of growth factors or the transplantation of genes that produce growth factors have had limited success, with some side effects.

Mandel's research group has concentrated on using an adeno-associated virus to engineer brain cells in animal models with genes that can protect dopamine-producing cells, which then do the vital work of producing GDNF, short for glial cell line-derived neurotrophic factor. The naturally occurring protein is important for the survival of dopamine-producing neurons during brain development, and a survival factor when given to adults.

In this case, scientists engineered the virus with two genes that must act in concert to produce the protein. But this precise interaction can be inhibited with dietary doxycycline, an antibiotic that is often prescribed in low doses to treat bacterial growth related to acne.

Depending on the amount of the antibiotic, protein production can be reduced or stopped, which would for the first time give medical investigators the ability to regulate gene therapy after the treatment was delivered.

"With this technique, you could adjust the therapy in the patient," said Fredric P. Manfredsson, a postdoctoral associate in UF's department of neuroscience. "That would be extremely helpful because no one is really certain yet what dosage is required for a protective effect in humans. The process is also much more sensitive than we had imagined it would be. GDNF production can be shut down completely with a dose of doxycycline that is much smaller than what is commonly prescribed."

Mandel said that adding the safety construct to the gene vector and proving its effectiveness were arduous tasks in which Manfredsson played an essential role.

A variety of methods were used to gauge GDNF production, but one was uncommon and involved the novel observation of the rats' weight. In prior research, the scientists had found delivering the protein to certain regions of the brain would hinder weight gain in younger rats and can cause weight loss in older rats. In the recent experiments, scientists found they could control the rate of weight gain in the rats with dietary doxycycline, which essentially verified they were controlling the GDNF therapy.

"The ability to control gene regulation is important for the future development of gene therapy, and optimizing its safe application to humans," said Dr. Mark Tuszynski, a professor of neurosciences and director of the Center for Neural Repair at the University of California, San Diego, who did not participate in the research. "The work of Dr. Mandel and colleagues brings us an important step closer to this goal."

Rheumatism Video Discloses Center Of Inflammation At An Early Stage

ScienceDaily (Aug. 29, 2009) — It can strike anyone: rheumatism occurs just as often as diabetes, arteriosclerosis and cancer combined. Approximately one percent of the population is stricken with the rheumatoid arthritis. Mostly it begins with initial inflammation in the finger joints. If it is discovered in time and a treatment is begun, the chances are good that the dreaded joint damage can be averted.

Now scientists at the Physikalisch-Technische Bundesanstalt (PTB), Berlin Institute, in a cooperation project with several partners, have developed such an early detection method. Their optical imaging method for rheumatism works with a fluorescent dye which is stimulated by near-infrared light and is absolutely harmless. With this method, centres of rheumatism can be detected at an earlier stage than is possible with an X-ray apparatus and is less expensive than a magnetic resonance tomograph.

Following the promising results of a clinical preliminary study still ongoing, the Berlin medical technology company "mivenion" acquired licensing rights from PTB in order to prepare a larger study. Also, the statutory health insurance companies have already shown interest in the imaging method, which should make rheumatism diagnostics and treatment distinctly more cost effective – particularly as a new, specific rheumatism contrast medium could in future improve it still another step.

HIV Subtype Linked To Increased Likelihood For Dementia

ScienceDaily (Aug. 29, 2009) — Patients infected with a particular subtype of HIV, the virus that causes AIDS, are more likely to develop dementia than patients with other subtypes, a study led by Johns Hopkins researchers shows. The finding, reported in the September Clinical Infectious Diseases, is the first to demonstrate that the specific type of HIV has any effect on cognitive impairment, one of the most common complications of uncontrolled HIV infection.

HIV occurs in multiple forms, distinguished by small differences in the virus' genetic sequence and designated by letters A through K. Certain subtypes appear to cluster in particular areas of the world, and others have been associated with different rates of progression to full blown AIDS. Of the 35 million people living worldwide with HIV, the majority live in sub-Saharan Africa, where subtypes A, C and D dominate.

Nearly half of patients with advanced HIV infections have at least mild cognitive impairments, and about 5 percent have the severe form of cognitive impairment known as dementia.

In earlier research, Ned Sacktor, M.D., and his colleagues found that about 31 percent of patients visiting an infectious disease clinic in the Ugandan capital, Kampala, where subtypes A and D dominate, had dementia. The finding led him and his team to wonder whether patients with different subtypes had different rates of dementia.

Sacktor, professor of neurology at the Johns Hopkins University School of Medicine and a clinician at the Johns Hopkins Bayview Medical Center, and his colleagues studied 60 HIV-infected patients from a Kampala clinic. All of the subjects had been part of a different study testing the effect of anti-retroviral drugs on cognitive impairment, but had not begun taking the drugs. After determining each patient's HIV subtype, they performed a battery of neurological and cognitive tests to assess each patient's brain function.

As expected, the majority of the patients had HIV subtypes A or D. Out of the 33 subtype A patients, the researchers determined that seven had dementia, or about 24 percent. However, out of the nine patients with subtype D, 8 had dementia, about 89 percent.

"We were amazed to see such a dramatic difference in dementia frequencies between these two subtypes," Sacktor says. "If this is the case in all of sub-Saharan Africa, HIV-associated dementia may be one of the most common, but thus far unrecognized, dementias worldwide."

The research suggests that some biological property of each subtype seems to influence the likelihood that infected patients will develop dementia, says Sacktor. He and his team hypothesize that subtype D may cause more inflammation and injury in the brain, a possibility they are currently investigating.

Parasites Ready To Jump: Even DNA Is Subject To Attack By Parasites

ScienceDaily (Aug. 29, 2009) — Transposons are mobile genetic elements found in the hereditary material of humans and other organisms. They can replicate and the new copies can insert at novel sites in the genome. Because this threatens the whole organism, molecular mechanisms have evolved which can repress transposon activity. Professor Klaus Förstemann of the Gene Center of Ludwig-Maximilians-Universität (LMU) in Munich and a team of researchers working with the fruitfly Drosophila melanogaster have now uncovered a new type of cellular defence that acts against DNA sequences present in high copy numbers inside the cell, even if they have not integrated into the genome.

Small molecules of RNA (a class of nucleic acid closely related to the genetic material DNA) play the central role. "Transposons are genomic parasites, so to speak", says Förstemann. "If they are allowed to proliferate, the genome can become unstable or cancers can develop. We now want to find out whether mammalian cells possess this newly discovered defence mechanism and to elucidate precisely how it works."

Transposons constitute a significant fraction of the genomes of most higher organisms. Indeed, it is estimated that these mobile elements, which include one or more genes, make up as much as half of the genetic material. "This demonstrates", says Förstemann, "that it is not always possible to tame these "selfish" genetic elements, although highly efficient mechanisms of defence have evolved. For instance, in the germ cells, which are required for reproduction, the system of so-called piRNAs ensures that transposon activity is inhibited – but only if these RNAs are transmitted from the mother. Disruption of this system usually leads to a drastic reduction in the fertility of the progeny.

Germ cells are an ideal target for transposons, since these cells pass their genetic material – together with integrated mobile elements – on to all cells of the progeny. But normal body (somatic) cells can also be attacked by transposons. For example, certain viruses carry transposons in their genomes and introduce them into the host cells they infect. Therefore, transposon activity must also be repressed in somatic cells. Recently so-called endo-siRNAs that perform this function were discovered in the fruitfly. A similar class of molecules has been found in mice.

By means of a process called RNA interference, the siRNAs enable the cell to recognize and destroy messenger RNAs derived from transposons. The researchers in Förstemann's group were able to identify a protein that is essential for the production of endo-siRNAs. It turns out that this is a previously unknown variant of the protein "Loquacious". In Drosophila, Loquacious can bind to specific RNA molecules that serve as precursors of the endo-siRNAs. Furthermore, the team pinpointed an entirely novel feature of this system: Repression of transposon activity was also detectable when multiple copies of a mobile element were present in the cell but not yet incorporated into the genome.

The phenomenon of RNA interference first came to light only a short time ago, but has already become a well established field of study. Thanks to more recent findings, the known repertoire of small RNAs has grown. As Förstemann stresses, "It is therefore particularly important to discriminate between the various molecular classes in terms of their modes of synthesis and specific functions". This is no easy task, since all these molecules are similar in size and virtually indistinguishable chemically. "We will now test whether the mechanism we have found in Drosophila also exists in mammalian cells. We would also like to know how the mechanism is targeted specifically against sequences present in high copy numbers".

Surprising Results In Teen Study: Adolescent Risky Behavior May Signal Mature Brain

ScienceDaily (Aug. 26, 2009) — A long-standing theory of adolescent behavior has assumed that this delayed brain maturation is the cause of impulsive and dangerous decisions in adolescence. The new study, using a new form of brain imaging, calls into question this theory.

A new study using brain imaging to study teen behavior indicates that adolescents who engage in dangerous activities have frontal white matter tracts that are more adult in form than their more conservative peers.

The brain goes through a course of maturation during adolescence and does not reach its adult form until the mid-twenties. A long-standing theory of adolescent behavior has assumed that this delayed brain maturation is the cause of impulsive and dangerous decisions in adolescence. The new study, using a new form of brain imaging, calls into question this theory.

In order to better understand the relationship between high risk-taking and the brain's development, Emory University and Emory School of Medicine neuroscientists used a form of magnetic resonance imaging (MRI) called diffusion tensor imaging (DTI) to measure structural changes in white matter in the brain. The study's findings are published in the Aug. 26, 2009 PLoS One.

"In the past, studies have focused on the pattern of gray matter density from childhood to early adulthood, says Gregory Berns, MD, PhD, principal investigator and professor of Psychiatry and Neuroeconomics at Emory University and director of the Center for Neuropolicy. "With new technology, we were able to develop the first study looking at how development of white matter relates to activities in the real world."

Gray matter is the part of the brain made up of neurons, while white matter connects neurons to each other. As the brain matures, white matter becomes denser and more organized. Gray matter and white matter follow different trajectories. Both are important for understanding brain function.

The study enrolled 91adolescents ages 12 through 18 over a three-year period. Levels of engagement in dangerous behaviors were measured by a survey that included questions about the teens' thrill seeking behaviors, reckless behaviors, rebellious behaviors and antisocial behaviors. DTI was used to measure corresponding structural changes in white matter.

"We were surprised to discover that risk-taking was associated with more highly-developed white matter – a more mature brain," says Berns. "We were also surprised to learn that except for slightly higher scores in risk-taking, there was no significant difference in the maturity of the white matter between males and females."

Berns suggests that doing adult-like activities requires sophisticated skills.

"Society is a lot different now than it was 100 years ago when teens were expected to go to work and raise a family," says Berns. "Now, adolescents aren't expected to act like adults until they are in their twenties, when they have finished their education and found a career.

"You could make the case that in this country, biological capacity shows up long before the wisdom that comes with time is fully developed," notes Berns.

Berns says more studies need to be done to determine if early brain development predisposes someone to engage in risky behaviors, or if the risky behavior drives the maturation of the brain.

The CDC reports that, 27,000 people between the ages of 10 and 24 die from bad decisions in the United States per year. Additionally, it has been shown that the period of mid-adolescence (ages 15 through 19) is the time when teens are more likely to begin high-risk behaviors such as drinking, abusing drugs or driving recklessly.
